Fire and Rescue NSW said the blaze broke out about 4.25am on Tuesday at a gym near the intersection with Stubbs Street, and up to 100 firefighters were at the scene fighting the blaze in the two-storey commercial building.
The cause of the fire was not immediately known.
Firefighters, some in cherrypickers, were still dousing the flames just after 7am, but the blaze had been contained. The gym was extensively damaged, but firefighters prevented the blaze from spreading to neighbouring buildings.
Crews were using fire hydrants from the opposite side of Parramatta Road and running hoses across the road.
Transport Management Centre spokesman Brett Moore initially advised motorists to avoid Parramatta Road at Auburn if possible, or expect extensive delays throughout the morning peak.
All lanes were initially closed in both directions, but re-opened just before 9am.
