Ambulance, police and police rescue responded to the accident at 9:20am.
A man in his 50s was trapped in his car after the vehicle ran off the road near Tiyces Lane, Boxers Creek.
The vehicle was travelling Northbound on the Hume Highway.
The man suffered a head injury and an injury to his left shoulder. He was transferred by helicopter to hospital in a serious condition.
Traffic in both directions has not been affected.
